Luminescence Technologies on the Hidex Sense
Just like LSC, luminescence recommends the use of white microplates in order to reflect light and maximise the light output signal. Without the need for any tools or manual handling the aperture can automatically be minimised within the software at a touch of a button which reduces cross-talk. The signal-to-background can also be optimised automatically by auto-focusing within the depth of the well.
Switching between top and bottom read is also made easy within the Hidex Sense. Real time bottom read can also be done through the use of injectors used for fast kinetic assays.
The New NanoBRET Platform Advances on from Bret Technology
Bioluminescence Resonance Energy Transfer (BRET) is a technique that has been used for many years to quantify dynamic interactions between proteins. However, the technique does have limited sensitivity and dynamic range which is a problem for certain assay types.
A new platform of BRET has now been developed called NanoBRET as a novel technology for analysing protein-protein reactions. This works via Nanoluc which is a bright luciferase to tag intracellular proteins with a long wavelength fluorophore (Halotag). Nanoluc is suited as an energy donor because of its narrow spectrum, high emission and small size, which ultimately produces greater light intensity and increased spectral resolution compared to standard BRET.
Direct Luminescence Is up to 10x More Sensitive
The photomultiplier tube (PMT) that is used for LSC can also be used to perform high sensitivity direct luminescence.
To perform this, the PMT manoeuvres into position and lowers down to press against the microplate.
This captures the emitted light in its entirety which reduces cross-talk and produces results up to 10x the sensitivity of standard luminescence.
A New Software Update Allows for Automatic Rate Calculations
Software on the Hidex Sense is constantly being developed and updated based on our user's needs. The latest update now includes automatic rate calculations such as Michaelis-Menten for enzyme kinetics.
This update reduces number crunching for the user, lowering the chance of human error and improving efficiency. This is done by automatically calculating the relationship between the substrate concentration and reaction rate.
